These slides come pre-treated with Calcofluor M2R, the preferred fluorescent probe for fungi. Here's how they are used:
Remove the protective top layer
Set your pipetter to 10µl
Add 10µl sample suspension to the center of the well
Use the pipette tip to spreasd the sample throughout the well
Dry at 42C for 5min
While the slide is drying set your pipetter to 7µl and assemble the immersion oil
Add 7µl immersion oil to the center of the dried test slide
Apply the coverslip, being careful to center it carefully
Press gently to seal the coverslip well. Decontaminate as per SOP.
Observe using UV Excitation
Yeast bud scars fluoresce bright Blue/White when probed with Calcofluor. This is a diagnostic test. No other cell fluoresces like this with Calcofluor
